Tg(Sox10-icre/ERT2)2Ldim

The bacterial artificial chromosome (BAC) RP23-28011 containing the Sox10 locus was modified to construct a transgene. A targeting vector containing the sequence for a codon improved cre recombinase (icre) fused cDNA encoding mutated estrogen receptor (ERT2), SV40 polyA sequence and an Frt-flanked PGK-neo cassette replaced exon 3 of the Sox10 gene in the BAC by homologous recombination in bacterial cells. Arabinose-induced Flpe expression removed the neo cassette. The purified transgene was microinjected into C57BL/6 oocyte pronuclei. Two founder lines, 1-Sox10 and 2-Sox10, were obtained. Line 1 was determined to have 2 copies of the transgene, and line 2, which showed higher expression, contained 9 copies. (J:185481)
