A 4.2 kb fragment of the mouse Nphs1 (nephrin) promoter was cloned upstream of the optimized Flp recombinase (Flpo) with a 3' bovine growth hormone polyadenylation sequence. The construct was microinjected into fertilized pronuclei. 18 founders were obtained with 3 demonstrating excision of the Frt-flanked neo from the Lama5neo allele. Flpo mRNA was detected by in situ hybridization in all three lines (1,3,5). Lines 1 and 3 when crossed to Lama5tm3Jhm mice showed higher podocyte expression of FLPo and normal tubular basement membrane levels of laminin alpha-5 (Lama5). (J:185850)
