Mouse fertilized oocytes were microinjected with transgene constructs in which the human MHC class II genes HLA-DRA and HLA-DRB1*0405, whose protein products together constitute HLA-DR4, reside downstream of the promoter region (nt -2172 to +12) of the mouse MHC class II Ea gene. The HLA coding sequence, in each case, is inserted into a ~1200 bp segment of the rabbit beta-globin gene which provides an intron, 3'-untranslated region (3'-UTR) and polyadenylation signal.
