A construct containing an eGFP gene and an inverted neo cassette was inserted in frame in exon 2 after the initiation codon via homologous recombination. RT-PCR and western blot analysis confirmed the absence of expression in the brains of homozygous mice. EGFP expression was detected by RT-PCR and live fluorescent imaging in the nervous system and liver. (J:184856)
