A construct in which the 4.6 kb porcine SRY promoter drives expression of the cre recombinase sequence was microinjected into male pronuclei of single-celled FVB/N embryos. A second construct encoding a tyrosinase minigene was co-injected to facilitate identification of transgenic animals. Pigmented founders were bred to wild-type FVB/N mice to ensure single integration sites. Three founder lines were obtained and crossed to cre reporter ROSA-YFP mice to analyze cre activity in newborn tissues from double transgenic mice. The three lines displayed similar results and one line was propagated for further studies. (J:184604)
