Exon 16 was replaced with an in frame RFP cassette and an FRT flanked neo cassette via homologous recombination. This disrupts the Dido3 isoform while leaving the other 2 isoforms intact. RT-PCR confirmed the absence of expression of isoform 3 and the presence of isoforms 1 and 2 in homozygous embryos. (J:183346)
