The cre coding sequence was inserted in-frame into exon 2 of the Phox2b gene in BAC RP24-95M11, so that a fusion protein contain the first 82 amino acids of Phox2b fused to cre recombinase will be produced. A two-step recombineering strategy was used with the first step resulting in the introduction of a dual antibiotic resistance cassette into exon 2 by homologous recombination and selection of positive clones, followed by a second round of recombination that exchanged the resistance cassette with one contain cre and an ampicillin resistance cassette. Correctly targeted BAC clones were further modified by removal of the loxP and loxP511 sites in the backbone so that undesired cre recombination would not occur. The purified BAC was injected into fertilized eggs to produce to founder lines with one being analyzed in detail. (J:180432)
