This transgene contains the regulatory sequences of the mouse enamelin (Enam) gene driving expression of the E. coli lacZ coding sequence modified to encode beta-galactosidase (beta-gal) with an amino-terminal nuclear localization signal (NLS). The mouse sequence extends from 4, 485 bp upstream of the Enam transcription initiation codon through the first 24 bp of exon 3, 5 bp upstream of the translation initiation codon. Beta-gal activity in transgenic mice fairly recapitulates the spatiotemporal expression of endogenous mouse Enam mRNA, occurring in secretory stage ameloblasts from birth through eruption of the last molars on postnatal day 17.
