A floxed neo cassette was inserted upstream of exon 3 and a loxP site was inserted downstream of exon 4 via homologous recombination. Cre mediated recombination removed the neo cassette and exons 3 - 4. Western blot analysis of total retina lysate from homozygous mice confirmed the absence of the 69 kDa isoform but did detect low levels of expression at about 45 kDa. RT_PCR analysis also detected low levels of expression of a short transcript. Sequencing determined this transcript was formed by alternative splicing of exon 1 to exon 5. (J:177916)
