A bacterial artificial chromosome (BAC; RP24-85B20) was modified by homologous recombination to insert a codon-improved cre (icre) fused to a mutated human estrogen receptor ligand-binding domain (ERT2). The targeting construct, which also contained an Frt-flanked ampicillin resistance cassette, was electroporated into bacterial cells containing the BAC. Cells with recombinant BAC clones were induced with L-arabinose to induce Flp recombinase expression to excise the resistance marker. The BAC was isolated, purified and injected into mouse pronuclei to obtain transgenic animals. No founder information was available so line 1 was entered. (J:179050)
