A floxed neomycin selection cassette was inserted in "differentially methylated region 1" (DMR1) upstream of Igf2. Insertion was in HpaII site 4 located in a Lrrfip1 (GCF2) binding consensus sequence. Properly targeted mice were crossed to mice carrying Tg(Pgk1-cre)1Lni to excise the selection cassette and leave behind a single loxP site that disrupts the GCF2 binding element. (J:178292)
