A puromycin-deltaTK (truncated thymidine kinase) sequence was inserted into the Sox2 locus by homologous recombination in V6.5 ES cells, placing TK under control of endogenous regulatory elements. In the mouse line generated, exposure to gancyclovir (GCV) will result in death of Sox2-positive (TK-expressing) dividing cells. (J:177655)
