A loxP site was inserted 316bp downstream of exon 6 in intron 6. A loxP and Frt flanked neomycin selection cassette were inserted in intron 3. The neo selection cassette was subsequently removed by crossing to mice expressing flp recombinase. This left exons 4, 5, and 6 flanked by loxP sites. However, cre recombinase failed to excise the floxed exons in living mice although excision succeeded in cultured cells. (J:174970)
