A construct, which contained (5' to 3') an adenoviral splice acceptor site (SA); a wild-type FRT site, a floxed inverted luciferase cassette; a puromycin N-acetyltransferase (PAC) gene; a non-integrating FRT site; and a promoter and start-codon deficient neomycin phosphotransferase gene, was inserted into the locus. This allele is suitable for RMCE to replace the inverted luciferase gene with a gene of interest. (J:173806)
