A BAC clone (RP24-148N) was modified by homologous recombination to generate an inducible cre transgene. The cDNA encoding tamoxifen-inducible cre recombinase (cre/ERT) was inserted after the 5' UTR, followed by a rabbit beta-globin poly adenylation sequence. The linearized 150kb construct was injected into pronuclei of 1- or 2-cell stage nuclei from B6SJL mice. Four founders were obtained and crossed to Z/EG reporter animals to assay EGFP expression in offspring. Line 467 displayed the highest cre efficiency and was chosen for further studies. (J:167748)
