This transgene encodes an immunoglobulin heavy chain (IgH) whose variable region is that of the monoclonal antibody expressed by the hybridoma cell line 6H4, which recognizes the normal endogenous mouse prion protein (PRNP); this is joined to the IgH-mu constant region (Cu) of the IgHa haplotype. Transgenic mice develop high titres of anti-PRNP antibodies composed of transgene-derived IgMa mu heavy chains paired with various endogenously encoded immunoglobulin kappa and lambda light chains, but have no detectable IgHM/D from endogenous genes.
