The firefly luciferase gene (from the pGL2 plasmid, Promega) was introduced into the ROSA26 locus using a knock-in strategy. The targeting vector was constructed to contain the coding sequence of firefly luciferase downstream of a loxP-flanked STOP cassette (consisting of a promoter-less neomycin gene followed by a three SV40 polyadenylation sequences).
