A lacZ/neomycin cassette was targeted at exon 3 (the first coding exon) by homologous recombination. Exon 3 is partially deleted which generates a premature stop codon. qRT-PCR and Western blot analysis showed that RNA and protein levels were drastically reduced in the hippocampus.
