A 540 bp region including the entire first coding exon was deleted and an IRES-hAP and floxed PGK-neo cassette was inserted in exon 2 via homologous recombination. Western blot analysis confirmed the absence of protein expression in homozygous embryos. Alkaline phosphatase activity mirrors endogenous gene expression. (J:171767)
