A targeting vector was constructed to replace part of exon 2 of the Gpr55 gene (containing the entire coding region) with a selection cassette. Homologous recombination was carried out in 129SvEvBrd-derived ES cells, followed by injection of targeted ES cells into blastocyts. Loss of Gpr55 mRNA was confirmed by RT-PCR which demonsrated lack of Gpr55 cDNAs in homozygous animals. (J:171771)
