Tg(Sox10-GFP,-DTA)1Wdr

A 120 kb fragment of a genomic PAC containing the entire Sox10 locus was used to generate the transgene. The region spanning exons 3-5 of Sox10 was replaced by homologous recombination in bacteria with a loxP-flanked eGFP polyA cassette followed by sequences encoding an attenuated G383A version of the diphtheria toxin, subunit A (DTA). An Frt-flanked Cm cassette was included for selection purposes, and was removed by transient activation of Flp in bacterial cells. In presence of cre recombinase, the reporter cassette is excised, allowing expression of DTA and ablation of the expressing cells. (J:105334)
