The luciferase gene, followed by SV40 poly(A) signal sequence, and a loxP site flanked neomycin resistance gene cassette, was inserted into exon 23 before the stop codon, in-frame with the targeted gene coding region. The inserted sequence replaces the endogenous 3' UTR. This creates an allele coding for a chimeric peptide of the endogenous protein fused at the C-terminus to the luminescent marker. (J:171738)
