A lacZ and FRT flanked neo cassette was inserted immediately downstream of the transcription start site via homologous recombination. Flp mediated recombination removed the neo cassette. Quantitative PCR analysis indicates that expression of the short transcript is decreased to about 6% of wild-type and expression of the long transcript is absent in MEFs from homozygous mice.
