A genomic fragment of the murine Aqp2 gene in a PAC was modified by homologous recombination in bacterias to put the sequence for a cre/ERT2 fusion protein at the ATG site of the Aqp2 locus. This construct was microinjected into FVB/N pronuclei to produce transgenic animals. 3 lines with 1, 4, and 9 copies of the transgene were established and analyzed for cre expression. The lines show copy number-dependent quantitative differences but have identical expression profiles. The line with 4 copies was chosen for further studies. (J:169670)
