Exons 2 and 3, encoding the first two EGF-like domains, were replaced with a lacZ and neo cassette. Spatiotemporal expression of lacZ, as determined by X-gal staining of whole and sections of embryos, is reported to resemble closely that previously reported for mRNA expression from the wild-type gene. Western blot analysis detects a truncated protein in homozygous mutant mice. Wholemount immunohistochemical analysis employing an antibody to an epitope in the spacer region, which is encoded by sequence downstream of the cassette insertion site, stains membranes of wild-type, but not of mutant epithelial cells.
