A FRT-flanked PGK-neo selection cassette resides downstream of the polyadenylation signal of the endogenous gene, and loxP sites are present 46 bp upstream of the ATG translation initiation codon and downstream of the neo cassette. The 3' loxP site is followed by the coding sequence for E. coli beta-galactosidase (lacZ) with a nuclear localization signal. (J:164344)
