A loxP site was inserted upstream of exon 3. A cassette, which contains (5' to 3') a loxP site, FRT site, splice acceptor, IRES, beta-geo cassette, stop codon, polyadenylation sequence, and FRT site was inserted downstream of exon 4. The inserted sequence disrupts expression of endogenous Grn and instead expresses a lacZ. Immunohistochemistry and western blot analysis confirmed the lack of protein expression in the brain.
