The first intron, encoding the transactivation domain and the first of the protein's three zinc fingers, has been replaced with a neomycin resistance cassette. Reverse transcription followed by PCR (RT-PCR) of mRNA from hearts of homozygous mutant mice detects no transcripts from the targeted gene; heterozygous mice express approximately half the wild-type level of mRNA.
