A 4.7 kb BamHI-HindIII genomic segment containing the first coding exon was replaced by a floxed neomycin selection cassette. The translation start site and part of a homeodomain were removed. The Prm1-cre transgene present in PC3 ES cells excised the selection cassette when it was expressed in spermatids of male chimeras. (J:164796)
