A floxed neo cassette was inserted upstream of exon 9 and an additional loxP site was inserted downstream of exon 9. Cre mediated recombination removed the neo cassette and exon 9. This exon codes for an essential catalytic motif of the deubiquitination domain. The absence of protein expression was confirmed by western blot analysis on fibroblast extracts. (J:162840)
