A 3.6 kb fragment including exon which encodes for the entire open reading frame was replaced with a tau-lacZ reporter and a self-excising neo cassette via homologous recombination. In situ hybridization confirmed the absence of transcript in brains from homozygous mice. X-gal staining confirmed that lacZ expression recapitulated endogenous gene expression. (J:161776)
