A cre-polyA-FRT-ampicillin-FRT targeting cassette with Slc2a2 homology fragments and a nuclear localization signal (nls) was inserted by homologous recombination at the ATG site of the Slc2a2 (Glut2) gene in EL250 bacteria. The FRT-ampicillin-FRT cassette was then removed in the presence of arabinose. Six line were obtained and crossed to reporter mice to verify cre activity. Two of these lines gave appropriate expression in pancreatic beta cells, liver, and intestine. Cre activity in the brain was in agreement with reported Slc2a2 expression.
