An EGFP construct was inserted in frame with the ATG start site followed by a floxed neomycin selection cassette. This was transfected into PC3 ES cells containing th Tg(Prm-cre)70Og transgene. The transgene is only active in male germline cells. Neomycin is thus excised in the germline. No gene expression was detected in the liver by PCR and no protein product was found in serum by Western blot. (J:159600)
