The inserted construct contains a "CAG promoter," which consists of the cytomegalovirus immediate early (CMV-EI) enhancer followed by a 1.3-kb DNA segment including the promoter, first exon and first intron of the chicken beta-actin gene, with the 3' splice junction sequence replaced by that of the rabbit hemoglobin beta gene, driving ubiquitous expression of the mouse cDNA with nucleotide substitutions that results in the amino acid substitution of histidine for arginine at position 172 (R172H). The construct is preceded by a floxed STOP cassette and followed by an IRES-EGFP. The R172H mutation is predicted to create a dominant-negative product that is only expressed following cre mediated recombination to remove the STOP cassette. (J:158953)
