A loxP-flanked neomycin resistance cassette was introduced to intron 4 and an additional loxP site was placed in intron 3. Cre-mediated recombination resulted in germline deletion of exon 4 and the neomycin cassette. The absence of protein expression was confirmed by western blot analysis on kidney and spleen extracts.
