A vector composed a 5' fragment of Flt4, an EGFP cDNA with a Pgk polyA signal sequence followed by an FRT-flanked Pgk-gb2-neo cassette ligated upstream of the first ATG of the Flt4 gene, and a 3' homology arm encoding an intronic region between exons 1 and 2 of Flt4 was used for targeting the Flt4 locus in (C57BL/6J x 129/SvEv)F1 ES cells. Exon 1 was replaced by EGFP after homologous recombination. The neo cassette was deleted in heterozygous mice by crossing with FLP66 deleter mice. No Flt4 protein is detected by Western blot in homozygous embryos. (J:159024)
