A targeting vector composed of a 1.2 kb fragment of murine Nkx6-2 containing Ret exons 12 and 13 flanked by loxP sites upstream of an frt-flanked neomycin cassette was generated from a BAC clone containing the Ret gene. The vector was incorporated into E14 ES cells for homologous recombination. Two indepent recombined clones were identified and germline transmission was verified by PCR. (J:131770)
