LoxP sites were inserted into exon 1, 167 nucleotides upstream of the start codon, and downstream of this exon. This exon encodes a short 5' UTR and 40 amino acids of the 356 including the signal peptide. Cre mediated recombination removed the selction cassette leaving exon 1 floxed. (J:158299)
