The coding sequence for cre recombinase, followed by an IRES and the DsRed2 sequence was placed downstream of a 3kb cholecystokinin (CCK) promoter. The construct was linearized, purified, and microinjected into the pronuclei of one-cell C57BL/6 embryos. Lines A, B, and C were established and analyzed. None of the three lines showed any observable DsRed fluorescence. Line C showed high levels of cre expression in the cortex, in a manner qualitatively similar to observed patterns of endogenous Cck mRNA expression, but showed almost no cre-mediated LacZ expression in any subcortical regions.
