A loxP-flanked neomycin cassette was inserted into the intron right before the exon encoding the C-terminus region of the targeted gene. An artificial stop codon plus an additional AatII restriction enzyme site were introduced by PCR mutagenesis so as to eliminate the last seven amino acids from the translated protein. The neomycin resistance cassette was excised in the germline by Cre recombinase. (J:183900)
