Exon 8 was replaced with a floxed neo cassette and a modified exon 8 that contains a C to T transition producing a termination codon instead of glutamine at position 307 (Q307X), modeling genetic lesions observed in patients with leukemia and myeloproliferative disorders. Cre-mediated excision driven by the Tg(Prm-cre)70Og transgene carried by the parental PC3 ES cells removed the neo cassette upon transmission from male chimeras. (J:157660)
