A mutated NLS1 with 5 amino acid substitutions (Lys to Gln) was introduced into exon 11 and a floxed neo cassette was introduced into intron 10 via homologous recombination. The neo cassette was then removed by Cre mediated recombination. Western blot analysis detected normal expression levels of the full length mutant protein in homozygous MEFs. Nucleo-cytoplasmic distribution of the mutant protein in homozygous MEFs is similar to that in wild-type cells. (J:156743)
