The transgene includes the human beta-actin promoter blunted at position -70 relative to the actin ATG start site. This is followed by intron 2 and part of exon 3 from rabbit beta-globin. The lacI coding sequence uses "synlacI" (which has been re-encoded to correspond to mammalian codon usage.) However, the 36 bp at the 5' end of the coding sequence are derived from the original bacterial gene as are the 150 bp between the 5' EcoRV site and the 3' PvuII site. The lacI coding sequence, with a nuclear localization signal appended to the carboxy terminal, is followed by exon 3 and intron 3 of rabbit beta-globin. This complex construct was optimized for ubiquitous lacI expression. Of three lines produced, 1 and 3 were widely expressed and line 13 had more limited expression. (J:153570, J:153571)
