Exon 23 was floxed and an FRT-flanked neo cassette was inserted into intron 23 via homologous recombination. The selection cassette and exon 23 were removed by flp and cre mediated recombination, respectively. Cre-mediated recombination results in out-of-frame splicing and nonsense mediated decay of the mutant transcript. (J:155739)
