Exons 23 and 24 were floxed and an FRT-flanked neo cassette inserted into intron 24 via homologous recombination. The selection cassette was then removed by flp mediated recombination. Cre-mediated recombination results in out-of-frame splicing and nonsense mediated decay of the mutant transcript. (J:155931)
