A floxed and FRT flanked PGK-neo cassette was inserted in intron2and an additional loxP site was inserted 5' of exon 2. The targeted region, encompassing exon 2, is 1.7 Kb in size. The FRT flanked PGK-neo cassette was excised by crossing to mice carrying the Tg(ACTFLPe)9205Dym transgene. Exon 2 was then deleted by crossing to mice carrying Tg(CMV-cre)1Cgn. Normal mRNA was not detected by Northern blot or RT-PCR. Protein could not be detected by Western blot. (J:155250)
