A transgenic construct was created with the Pgk1 promoter driving mouse codon-optimized FLP (FLPo) recombinase. The linearized construct was injected into fertilized C57BL/6 x C3H oocytes. Six founder mice showing germline transmission were generated. These animals demostrated FLP expression as early as 3.5 embryonic days and adults displayed ubiquitious expression in all tissues examined with strongest expression in the testis and ovary. Line 10 showed 100% recombination efficiency when crossed to mice expressing a Ghr allele with a FRT-flanked PGK-Neo cassette in intron 5.
