A targeting vector generated by bacterial recombineering replaced most of the Hes1 open reading from with a cre/ERT2 sequence linked to an Frt-flanked neomycin resistance cassette. Following homologous recombination in R1 ES cells, germline chimeras were generated and subsequently the neo cassette was excised in vivo by crossing mutants to Gt(ROSA)26Sortm1(FLP1)Dym mice.
