A CMV promoter drives ubiquitous expression of a double-stranded RNA construct consisting of a 498 bp anti-sense and sense fragment of Pqbp1 arranged tail to tail preceded by a ribozyme cassette and followed by a MAZ site for RNA Pol II pausing. Two lines were created (lines 1 and 2). Western blot analysis confirmed that both lines exhibit a 50% reduction in endogenous PQBP1 expression without affecting expression of GAPDH, alpha-Tubulin, beta-Actin, GFAP, or MTAP2.
