The targeting vector contains a diphtheria toxin cassette, a neo cassette in reverse orientation inserted 3' to exon 4 and loxP sites flanking the neo cassette and exon 4. Exon 4 includes coding sequences for the transmembrane regions, the ubiquitination site and zinc finger binding domain. Mice were crossed with mice carrying Tg(CAG-cre)12Miya to remove exon 4 and the neo cassette leaving a single loxP site downstream of exon 3. Protein was not detectable by Western blot and immunochemistry analyses. (J:154853)
